Malisse, who missed most of last season with a wrist injury, faces the 12th seed in Centre Court's second match.
Anne Keothavong opens on Centre Court against Venus Williams, with compatriot Chris Eaton, 661 in the world, third on Court One against Dmitry Tursunov.
Rafael Nadal and Maria Sharapova are also on Court One, while Andy Roddick completes the Centre Court line-up.
In facing the defending champion, Keothavong follows in the footsteps of compatriot Naomi Cavaday, who performed creditably by taking the first set into a tie-break in their first-round match on Tuesday.
"I'm sure I'll speak to Naomi about it," said the 24-year-old Londoner.
"I'm really excited about it. It's kind of what you play tennis for, to play these big matches."
But Britain's more realistic hopes of a representative in the third round lie with Murray, who put in a composed performance to see off Fabrice Santoro in round one.
However, the world number 11 is wary of over-confidence.
"Malisse is really talented," said the Scot.
"He's had quite a lot of problems with injuries over the last couple of years, but I practiced with him quite a lot at (coach Nick) Bollettieri's academy. He can play very well on grass.
"He probably isn't playing as well as he was when he made the semis, but he's a dangerous opponent."
Belgian Malisse once beat Greg Rusedski on Centre Court and says he is not worried about playing in front of the British crowd.
"Of course Andy will be favourite and he'll have the whole country behind him, but I'm used to that situation," he said.
"Once you're into the match, it's a great atmosphere. But once you're on the court you want to win and that's the only goal I'll have in my mind."
Eaton takes on 25th seed Tursunov following his first-round win against Serbia's Boris Pashanski. "I have watched him play on TV before so I know what to expect," said the 20-year-old.
Nadal, hoping to reach the final for the third year running, faces a potentially dangerous opponent in Latvian Ernests Gulbis.
Gulbis, 19, reached the quarter-finals at the French Open and followed that with a run to the third round at Queen's where he lost in a third-set decider to Murray.
Elsewhere Roddick faces talented Serb Janko Tipsarevic third on Centre Court. Tipsarevic caused a sensation at this year's Australian Open when he took Roger Federer to five sets.
In the women's draw, third seed and ex-champion Maria Sharapova meets fellow Russian Alla Kudryavtseva in the second match on Court One.
World number two Jelena Jankovic faces Spain's Carla Suarez-Navarro in the third match on Court Two.
